表觀遺傳染色體修飾酶PCR芯片可用于研究與基因組DNA和調(diào)節(jié)染色體可塑性組蛋白修飾有關(guān)的84個關(guān)鍵基因的表達,這些基因包括已知的或推測的。維持甲基化的DNA甲基轉(zhuǎn)移酶,負(fù)責(zé)CpG雙核苷脫甲基酶,催化組蛋白乙?;?、甲基化、磷酸化、泛素化的酶以及去乙酰酶和去甲基化酶都在此芯片上。該芯片還分析編碼SET編碼域的蛋白,他們都含有一個共同的編碼域,用以闡述對一些家族成員的組蛋白甲基轉(zhuǎn)移酶活性。在干細胞的發(fā)育晚期成為分化細胞,基因表達改變的發(fā)生可以用來調(diào)控染色體動力學(xué)。腫瘤細胞也展現(xiàn)出相對于正常細胞不同的基因表達譜,這表明在腫瘤生成中染色質(zhì)修飾和重構(gòu)的作用。通過實時定量PCR的方法,研究者即能夠利用該芯片簡單可靠地同時檢測與表觀遺傳染色體修飾酶有關(guān)的基因表達。 DNA Methyltransferases: DNMT1, DNMT3A, DNMT3B.
